COURSE CONTENT
Basic principles of spectroscopy. Light and its interaction with molecules.
The UV-VIS absorption: theoretical basis and applications. Correlation between molecular structure and optical properties. The chromophore. Role of experimental conditions on the optical properties of a chromophore. Chromophores in biomolecules. The Lambert-Beer law. Spectrophotometer. The use of UV-VIS spectroscopy in quantitative analysis.
Emission spectroscopy: theoretical basis of fluorescence and its applications. The fluorophores. Protein fluorescence of the proteins and the effect of temperature and solvent. Conformational stability and allosteric effects in proteins. Fluorescent probes for proteins and nucleic acids. Fluorescence in quantitative analysis. Fluorescence quenching and fluorescence energy transfer.
Chromatography: basic principles and applications. Theoretical basis of electrophoresis. DNA and protein electrophoresis. Effect of complex formation on the electrophoretic mobility of biomolecules.
Comparison of different methodological approaches to quantitatively describe drug-biomolecules interactions.
LEARNING OUTCOME
The course is divided in a theoretical (2 CFU) and in a practical (3 CFU) part. The two sessions are interconnected to offer the basic knowledge required to perform a qualitative and quantitative characterization of naturally occurring biomolecules (proteins, polysaccharides and nucleic acids). In particular the course will introduce to spectrometric and chromatographic techniques used to detect, identify and quantitatively determine the interaction between biomolecules and drugs. At the and of the course, students are expected to be able to evaluate analytical data and to critically discuss experimental details connected to the proposed experimental sessions. Additionally, they should be able to choose among different strategies for analytical research of drug-target interactions accordingly with the nature of the components of the system under investigation.
